I've heard around the forums that what squadmates you take with you on a mission depends on the mission itself. The problem is that while everyone is very willing to parrot this advice, no-one seems to be willing to expand on it. I'm hoping someone will be willing to do so, because vague and nebulous advice is never any good.

Geth/Mechs -- Zaeed for Disruptor Ammo

Husks -- Samara for Area Reave/Pull Field, Grunt or Jacob

After that it doesn't really matter. You can take anyone you want and they will work just fine. There are probably a few characters that are slightly more effective than others in certain areas, but it's not necessary to bring them along. Take who you like.

Still, I highly reccommend bringing Zaeed along for Geth missions unless you have your own Disruptor Ammo.

I'm a Soldier, so I'm pretty much set for Ammo. I generally bring Miranda (all my damage is from weapons, and she boosts weapon damage), then switch between Garrus (when going against Mechs and Blue Suns) and Grunt (for everything else).

Usually, you want a team that can fill the gaps in your own weaknesses. unfortunately, some of your teammates like Tali are best left up to specific missions.

Then I think you're set.

Don't sweat the details, just play however you like. This game is pretty forgiving.

Mordin is very good vs Krogans and Husks with maxed out incinerate blast and frozen blast. his frozen blast has only 4.5 sec cool down, best of any of your squad members, i suspect it's unintentional.

Miranda + Garrus/Grunt is a good team.

.

In my last playthrough as a Solider, I had no problens with defenses. So I actually maximized MIranda's Slam and brought another teammamte that could make massive damages do unprotected enemys.

.

Agains Husks, you can take Mordin and loyal Samara.

Yeah, on 4/5 of my playthroughs I used Miranda (romance) and Garrus (wingman) almost exclusively.

It works just fine.

Miranda- An all rounder, or for warp explosions.
Jacob- Squad Incendiary (for armour/cc/health regenerators), short range (reasonably tough to get into range), pull for warp explosions.
Zaeed- Squad Disruptor (mechs/shields/limited cc), Inferno Grenade (anti-armour (not great) and cc), Mid' and long range weaponry, Concussive shot for more cc.
Mordin- looow cooldown cryo power, Incendiary for excellent armour penetration/anti-regen, neuro shock for cc, reasonably tough with his passive boosts.
Garrus- Squad AP for excellent anti armour, Overload for anti-shields, Concussive for cc, mid/long range.
Grunt- Very, very hard to kill, short/mid range, rapes husks, squad incendiary for armour, concussive for limited cc.
Jack- Pull for biotic combos, squad warp ammo for barriers/armour/health, shockwave rapes on sub-hardcore, reasonable cooldowns.
Thane- Warp/throw for combos, long range (SMG is decent at short), passive grants ALOT of damage+, jury is still out on shredder ammo,
Samara- Pull for combos, Reave speaks for itself, mid range, good cooldowns.
Tali-Brutal against all kinds of synthetics.
Legion- WIDOW, also great against synthetics.

As you can see the best squaddies obviously depends on what you're fighting and what is equally important is what class Shepard is, as that can effect who to use for biotic combos, what ammo types to use, what ranges you're effective against and what defenses you're weak against.

The only class that actually got me through insanity was Shepard as an infiltrator and then Grunt and Miranda. I used them on all missions and got through without too much yelling in anger.
Miranda has the kickass Warp/Overload and then for Grunt I got all of his health upgrades and focused on making scanning a lot of planets so that I could buy pretty much every upgrade. It's slow going but it works.

As an adept, here is the basic vs guide:

1st choice is the usual optimal while 2nd choice is an alternate and equal in optimality as well.

vs. Geth:

1) Garrus and Tali (best choice - AI hack, 2 shield strippers, squad AP ammo)

2) Zaaed and Tali (for more of a shooting adept - AI hack, energy drain, squad disruptor ammo, inferno grenade)

vs. Blue Suns:

1) Garrus and Miranda : 2 overloads, squad AP ammo, warp detonation combo

2) Garrus and Tali (for more of an aggressive game): 2 shield strippers, AI hack for LOKI mechs, squad AP ammo, and drone for distraction

vs. Eclipse:

1) Garrus and Samara: overload, reave, concussive shot, pull/throw biotic combo, AP squad ammo

2) Miranda and Jack (for a more shooting adept): overload, pull/warp for biotic combo, shockwave to take people out of cover, squad warp ammo

3) Garrus and Thane (excellent sniper combo - expect a lot of headshots once you get someone stuck with singularity): overload, warp, throw, concussive shot, 2 sniper aimbots.

vs Blood pack:

1) Jacob and Mordin: squad incendiary ammo, pull for biotic combo, area incinerate, area cyro blast (fastest squadmate cc power)

vs Husks/Scions:

1) Jack and Mordin: squad warp ammo, fast pulls, area incinerate, area cyro blast (fastest squadmate cc power)

2) Jack and Samara: squad warp ammo, fast pull/throw, area reave

vs. Collectors:

1) Jack and Samara: squad warp ammo, fast pull/throw, area reave, shockwave to take enemies out of cover/push to ledges

2) Thane and Samara (works best if you get squad warp ammo as bonus skill): pull/throw for biotic combos, area reave, unstable warp

*NOTES:

Mordin's max cyro blast is the fastest cc power at 4.5 sec to recharge (comparable to Shep)

Jack and Samara have the fastest pull/throw/shockwave recharge after maxing their passive skills

I just finished Thane's Recruitment Mission, and I think I've found my second squaddie for Eclipse/Collector Missions. I'm not really a big fan of his weapon selection, but he's got Throw and Warp, which is pretty solid. And with his +50% Weapon Damage from his maxed Passive, he can dish out a lot of damage with his SMG. I have yet to give him a test run (just about to fire up the game and do it now), but on paper, it sounds solid. I just have to remember to save 9 Squad Points so I can yank the point out of Shredder Ammo once I do his loyalty mission.

Not a fan of his weapons? He uses a sniper rifle, that is the absolute best weapon for a squaddie to have. It has maximum range, maximum damage and maximum reload time (meaning he stays in cover and reloads most of the time).

True enough, but I generally like my squaddies to have Assault Rifles for general-purpose use.

My most used squad mates during my insanity run were: Tali, Garrus, Miranda and Thane.

Tali's drone was exceptionally usefull getting enemies to change their target away from me, and upgraded to Explosive Drone made it really usefull.

Miranda and Thane is the build i used during the Suicide Mission, both upgraded to heavy Warp. My Singularity + their Warps = a quiet Harbinger ;)

Had my share of wipes nonetheless.
