in the beginning of game I cannot open the locker to get the gun . The space key on my key board seems to have no effect. I have windows 7, maybe that is why I cannot open the locker. I am standing on the arrow notation but the space key will not open locker.

Probably solved it by now, but try saving then reloading, or exiting the game and reloading, and trying again. If that doesn't work, make sure the key is bound correctly, if its been changed then the hint will still tell you to use space. Other than that, i got nothing.
This also happens on a optional mission vs Blue Suns on a spacecraft (Gurnsbeck something??) in the last room everytime i do this mission, an upgrade is in a box and it *never* opens unless i save and reload. Took me a couple of playthroughs before google told me what to do and i got the upgrade at last.

As was the case for the two prior iterations of this exact same complaint (and still another dupe following this one), it is off topic here, as well as being cross-posted. Same deal for the newer dupe, with a different topic line. And, it has already been answered (caused by below minimum hardware).

That mean that none of the dupes were even "necessary" as bumps to the first one, and all should have been locked as SPAM, except that hardly any moerating of that nature has been enforced.

Over a time span of about five hours, averaging an hour and fifteen minutes apart, in two different off-topic forums (never did post anything in Tech, but a Mod has moved one of them there now), building up to an accusation that the prior posts has been deleted, and that there was a conspiracy to avoid admitting in the forums that Mass Effect was unable to run in Windows 7, he kept gradually becoming more strident.
So, yes, it will be a long time before this one (OP) catches on to how the real world actually works, if, in fact, he / she ever does.

Turn down resolution and effects and update DirectX and drivers.
Escape to the mission computer and then back to the game and press space immediately while looking at the box; you should have a brief opportunity to activate it.
