All voice actors for the voices, and some lookalikes for the real roles:
Meer for voice and Vanderloo for the Role forexample.

Except for Miranda who would just be played by Yvonne Strahovski <3

I don't think there should be a movie at all, because if you look at movies and games there not the best of friends, remember the Mario Bros. movie?

Katee Sackhoff as Shepard.

Or a new story altogether that doesn't mention Shepard at all as no matter what sex she's portrayed as, people are going to be peeved.

Maybe if this was Halo that might have been the case.Their is a Default shepard which every one who has played Mass effect knows. Why because he is on cover and in  the ads and if  you don't make any changes that is who you play with.

Thanks to the magic of scissors, whoever that guy is ain't on the cover of mine.
I don't know about you, but the Shepard I'm most familiar with (thanks to more than a few hours of playing the games) doesn't look like that person at all. Nor does she sound like he might (I live in Edmonton and have enjoyed Mark Meer's work on the stage several times, but have no idea what he's like in the games).

Anyway, on topic...

Maybe Lucy Lawless as Shepard.

10 years ago Lucy might have been able to pull it off. If anyone has seen the show Spartacus though I think she's sadly starting to show her age.

ME2 has just a little more cinematic potential than mario bros I reckon. *note understatement*

Oh, and there should be a prototype movie. Just thought I'd mention that.

Vin Diesel as Miranda

Vin Diesel as Kaiden

Vin Diesel as The Normandy

Vin Diesel as "space hampster"

Vin Diesel as Garrus

Vin Diesel as Tali

Vin Diesel as Jacob

Stephen Hawking as Commander Shepard

...in short, please don't make a Mass Effect Movie, it would suck no matter who you cast or who shot it. I love the games so far, but some film jackass would slaughter the concept.

P.S. Vin Diesel is dumb, or whatever his real name was before he changed it to "be cool."

I'm starting to hope that that is the case.

I'd like to see a feature-length movie or mini-series for ME, but using full CGI.
Probably shouldn't even feature/focus on Shepard (but all the characters from the games should have the same voice actors/actresses), so as not to generate a 'canon storyline' for both games. :)

If there's ever a Mass Effect film (hopefully there won't be) and I was tasked with assembling a standard Hollywood cast, it would consist of:

Commander Shepard - Jodie Foster
Let's face it, Shepard is automatically a far more interesting character if female.
Not only because of the socio-normative expectations, but because there's only really one precedent in the sci-fi genre; Sigourney Weaver's Ripley.
Jodie Foster is not only a commanding screen presence, but in my opinion one of the most capable film actors of the last 50 years. She would make an excellent protagonist.
PHOTO

David Anderson - Keith David (duh)
The man is a brilliant actor, with an equally amazing vocal authority.
A perfect choice.
PHOTO

Jeff "Joker" Moreau - Justin Theroux
I know I'm asking for trouble by saying this, but I think Seth Green is rather grating, with the exception of his work on Robot Chicken.
Theroux would be my replacement.
PHOTO

Ashley Williams - Asia Argento
While she has done her fair share of awful films, there's an inexplicable charisma to her.
My preliminary pick, although I must say I found this the hardest part to cast.
PHOTO

Kaidan Alenko - Henry Rollins
Alenko was so thoroughly devoid of interesting personal attributes that some contrasting casting is necessary.
Rollins would likely provide the 'oomph' necessary.
PHOTO

Garrus Vakarian - Brandon Keener (naturally)
Due to the alien physique of the turians, there's probably be up to some skilled puppeteering and other visual tricks to do Garrus justice.
Keener works perfectly fine in the games, so why not in the film?

Tali'Zorah nar Rayya - Liz Sroka (given)
Granted, I am unaware of how talented a physical actor Sroka is, but judging by her vocal work there would be enough gravitas to hold the part.

Liara T'Soni - Naomi Watts
One of the great sympathetic faces of modern cinema. And has proven in films such as Mulholland Drive and Funny Games that she can play vulnerable yet headstrong characters.
PHOTO

Urdnot Wrex - Ron Perlman
C'mon, what can you NOT cast Perlman as?
Easily one of the most versatile actors of the past thirty years; from The City of Lost Children to Hellboy a consistently great performer.

Doctor Chakwas - Helen Mirren
Enough said.
PHOTO

Donnel Udina - Robert Picardo
Simple. Nobody plays agitated beaurocrats like Picardo.
PHOTO

Saren Arterius - David Morrissey
A class act, and a class actor.

Matriarch Benezia - Ellen Barkin
Shares some features with Watts, and a fine thespian in her own right.
And even though she also shares Benezias more...err...'buoyant' attributes, I'd make sure there was no improbable cleavage on display. She deserves better.
PHOTO

------------MASS EFFECT 2------------

"The Illusive Man" - Tilda Swinton
I know it's an inherent contradiction, but bear with me.
In my opinion, TIM was neither intriguing nor enigmatic in the way ME2 presented him.
I would've preferred something more ambiguous; an utterly androgynous presence whose very appearance was a riddle in itself.
Swinton is a brilliant actor, and would make the part sing with nuance.
PHOTO

Jacob Taylor - Isaach De Bankolé
One of my favourite actors, and would likely infuse the otherwise terminally dull Jacob with plenty of edge and sex appeal.
PHOTO

Miranda Lawson - Selma Blair
Give her a proper costume and Blair would shine.
- But honestly, give her some real clothes.
PHOTO

Jack/Subject Zero - Rebecca Romijn
Watch Brian de Palma's "Femme Fatale".
You'll see.
PHOTO

Samara - Michelle Pfeiffer
I always though Samara had some of that classic golden age Hollywood to her.
You know, apart from the plunging neckline.
Pfeiffer is arguably one of the most talented performers in american motion pictures, and she definitely has a bearing worthy of the part.
PHOTO

Thane Krios - Jason Isaacs
One of my favourite choices. Isaacs would be perfect in the role.
PHOTO

Grunt - Steve Blum
Not a very interesting part to play, but Blum made an admirable effort in the game, so there you have it.

Mordin Solus - Michael Beattie
Nigh flawless in the game. No substitute necessary.

Legion - D.C. Douglas
Meh. It works.

Zaeed Massani - Bruce Campbell
"Groovay".
PHOTO

Kelly Chambers - Winona Ryder
I've always had a soft spot for Ryder. And I can definitely see her as the kind, affectionate yeoman.
PHOTO

EDI - David Bowie
Yes, yes it's another little gender game of mine.
So sue me. Subversive approach > conservative approach.

Morinth - Laura Harring
Unless we just had Pfeiffer reprise the role as her daughter with some superficial changes in the make-up, Harring would lend the part a fittingly eerie quality.

Megan Fox is awful choice for Jack... Although if she was in Jack's outfit I wouldn't mind what so ever. I like Natalie Portman. She is a good actress and surprisingly looks like Jack. She has the mouth from when we heard the Natalie Rap and she has the look from V for Vendetta.

Actually I would prefer a movie based on another part of ME history. First contact war anyone? Skylian Blitz?

Shepard's story would be great, but not so great as the story told on the game for sure.

I also have to say that Titan7771 made a very good list.

I was thinking of Jodie Foster too (and Sigourney Weaver for that matter), but wasn't sure if they'd be too old. Not that it isn't stopping some of the male Shepard choices. :)
I do have one small quibble with your comment here. Maybe as military trained, Ripley is a precedent, but for kicking ass? You can't forget Linda Hamilton in the Terminator movies! If age isn't a restriction, then she'd make a great choice too.

Kiefer Sutherland, Jack Bauer from 24. He pulls of a frikkin awesome voice as the Caller in Phone Booth. Don't know who he would fit, but man has he got a cool voice that could fit someone evil.

Well, my issue with Terminator is that they are such poorly written films.
But what ever my opinion of James Cameron may be, it's true that he's a sex-egalitarian filmmaker.
So yeah, I suppose you're right to an extent.
- Though Ripley is different simply due to the fact that she's the main character in all four Alien-films, whereas Schwarzenegger always had top billing in the Terminator franchise.

Megan Fox is a babe, but I don't think she could hold her own as Jack, namely because of her voice. Jack needs to be able to scream and be intimidating, Megan couldn't pull this off. Natalie Portman both looks like Jack, and has the intensity to pull her off. As for her height, directors are very good at making short actors look taller. Like Jason Statham, Tom Cruise, and Mark Wahlberg, all action stars, all stand at around 5 foot 6, hardly giants.
